Functional Genetic Diversity in American Mink (Neovison vison)
The release of domestic organisms to the wild is considered a threat to
biodiversity because the introduction of domestic genes through interbreeding can
negatively impact wild conspecifics via outbreeding and local extinction. In North
America, captive American mink (Neovison vison) are frequently escaping into the wild,
yet the impact of these events on the functional genetic diversity of wild mink populations
is unclear. I characterized domestic and wild mink in Ontario at 17 trinucleotide
microsatellites located in functional genes thought to be associated with traits affected by
domestication. I found low functional genetic diversity, as only 4 of 17 genes were
variable and of those four there was little evidence of allele frequency differences
between domestic and wild mink. Using redundancy analysis and a spatial analysis of
principal components on the four variable loci (AR, ATN1, IGF-1, and TOB1) I found no
evidence to suggest domestic release events are affecting functional genetic diversity of
free-ranging mink at the set of markers assessed.

Evaluating Environmental DNA (eDNA) Detection of Invasive Water Soldier (Stratiotes Aloides)
In 2008, the first North American water soldier (Stratiotes aloides) population was discovered in the Trent River, Ontario. Water soldier is an invasive aquatic plant with sharp, serrated leaves that has the potential to spread rapidly through dispersed vegetative fragments. Although it is too late to prevent water soldier establishment in the Trent River, its local distribution remains limited. In this study, environmental DNA (eDNA) was explored as a potential tool for early detection of water soldier. Species-specific markers were designed from chloroplast DNA regions matK and rbcL, and a qPCR assay with rbcL primers yielded the most sensitive detection of water soldier eDNA. Positive detections were obtained from six of 40 sampling locations, of which five were collected in Seymour Lake, an area with large patches of water soldier. As water soldier plants were known to be present at these sites, high eDNA concentrations were expected. The sixth positive detection from Trent Lock 5 (50 km downstream of Lake Seymour) was unexpected as it was obtained at a site with no water soldier sightings. This is one of the first studies to demonstrate the effectiveness of eDNA detection from aquatic plants.

Studies of the Giardia intestinalis trophozoite cell cycle
To study the Giardia intestinalis cell cycle, counterflow centrifugal elutriation (CCE) was used to separate an asynchronous trophozoite culture into fractions enriched for cells at the different stages of the cell cycle. For my first objective, I characterized the appearance of a third peak (Peak iii) in our flow cytometry analysis of the CCE fractions that initially suggested the presence of 16N cells that are either cysts or the result of endoreplication of Giardia trophozoites. I determined that this third peak consists of doublets of the 8N trophozoites at the G2 stage of the cell cycle that were not removed effectively by gating parameters used in the analysis of the flow cytometry data. In the second objective, I tested the use of a spike with RNA from the GS isolate of Giardia as an external normalizer in RT-qPCR on RNA from CCE fractions and encystation cultures of Giardia from the WB isolate. My results showed that the GS RNA spike is as effective as the use of previously characterized internal normalizer genes for these studies. For the third objective, I prepared two sets of elutriation samples for RNA seq analysis to determine the transcriptome of the Giardia trophozoite cell cycle. I confirmed the results of the cell cycle specific expression of several genes we had previously tested by RT-qPCR. Furthermore, our RNA-seq identified many genes in common with those identified from a microarray analysis of the Giardia cell cycle conducted by a collaborator. Finally, I observed an overall <4 fold change in differentially expressed genes during the G1/S and G2/M phase of the cell cycle. This is a modest change in gene expression compared to 10 - 30 fold changes for orthologous genes in mammalian cell cycles.

Building Individuals, Building the Economy: Labour Force Development in Canada's Territorial North
This thesis explores the neoliberal governmentality approach to education for Northern economic development that was prevalent from 2006 to 2015, during Stephen Harper's period as Prime Minister of Canada. Using a grounded theory approach, this thesis identifies three themes – Indigenous integration, education, and employment for labour force/ economic development – to direct an analysis on programs and funding supported by the Canadian Northern Economic Development Agency, Employment and Social Development Canada, and Indigenous and Northern Affairs Canada. This examination suggests that Federal programming and funding encouraged neoliberal governmentality approaches to Northern development and education. Specifically, the former Government interest in developing an Indigenous work force to serve labour market needs is brought to light.

Cytokinin biosynthesis, signaling and translocation during the formation of tumors in the Ustilago maydis-Zea mays pathosystem
Cytokinins (CKs) are hormones that promote cell division. During the formation of tumors in the Ustilago maydis-Zea mays pathosystem, the levels of CKs are elevated. Although CK levels are increased, the origins of these CKs have not been determined and it is unclear as to whether they promote the formation of tumors. To determine this, we measured the CK levels, identified CK biosynthetic genes as well as CK signaling genes and measured the transcript levels during pathogenesis. By correlating the transcript levels to the CK levels, our results suggest that increased biosynthesis and signaling of CKs occur in both organisms. The increase in CK biosynthesis by the pathosystem could lead to an increase in CK signaling via CK translocation and promote tumor formation. Taken together, these suggest that CK biosynthesis, signaling and translocation play a significant role during the formation of tumors in the Ustilago maydis-Zea mays pathosystem.

Aquatic Invertebrate Studies from Two Perspectives: Function (Shredders) and Distribution (Dytiscidae)
Leaf litter decomposition represents a major pathway for nutrient cycling and carbon flow in aquatic ecosystems, and macroinvertebrates play an important role in the processing of this material. To assess the causes of variable leaf breakdown and nutrient fluxes, I measured decomposition rates and the nutrient release ratios of decomposing leaf material across a broad latitudinal gradient in Ontario boreal lakes which varied in nutrients, temperature, and pH. I examined the effects of macroinvertebrates using inclusion and exclusion bags. Generally, leaves decomposed faster in nutrient-rich, warmer lakes. Macroinvertebrates increased decomposition rates but their effects were relatively small compared to regional effects of nutrients and temperature. In addition, we found differential effects of nutrients and temperature on nutrient release ratios, which were partially determined by the release and retention of N and P. These results indicate that changes in these important environmental lake variables could alter decomposition dynamics in Ontario lakes, with implications for nutrient cycling and the storage of this important external carbon source.
I studied the biogeography of predaceous diving beetles (Coleoptera: Dytiscidae) in two remote and understudied regions: the Far North of Ontario, and Akimiski Island, Nunavut. I identified 35 species from northern Ontario, including three first provincial records for Ontario, Acilius athabascae Larson (1975), Hygrotus unguicularis (Crotch 1874), and Nebrioporus depressus (Fabricius 1775). I also documented three significant range extensions and six gap-infills for this region. I collected and identified 16 species from Akimiski Island, Nunavut, which include several first time reports for these species for the Nunavut territory. My collections also extend the known ranges of five species into the Hudson Plains Ecozone. This work provides important baseline information on the distribution of diving beetles for these regions.

Archaeology, Engagement and Local Communities: The Stelida Naxos Archaeological Project and the Locals of Vivlos, Greece
This research is an ethnographic investigation into the relationships between the Stélida Naxos Archaeological Project and the local population of Vivlos, the region where the team takes their seasonal residence during their annual archaeological field season. Fieldwork in Vivlos revealed the local peoples' interest in archaeology, local legends, and Greek history. The people's cultural identity facilitated a sense of communal pride with hosting the archaeologists for their field season. The archaeologists' ethical considerations and their friendliness towards the locals during their time in Vivlos followed practices affiliated with public archaeology, laying the groundwork for maintaining positive working relations between the two groups.

Effects of flooding on nutrient budgets and ecosystem services
Increases in flooding due to anthropogenic influences such as climate change and reservoir creation will undoubtedly impact aquatic ecosystems, affecting physical, chemical, and biological processes. We used two approaches to study these impacts: a whole-ecosystem reservoir flooding experiment and a systematic literature review. In the whole-ecosystem experiment, we analyzed the impact of flooding on nutrient release from stored organic matter in an upland forest. We found that flooded organic matter produced N (nitrogen) and P (phosphorus), but that more N was released relative to P, increasing the N:P ratio over time. In the systematic literature review, we linked small (<10 year recurrence interval) and extreme (>100 year recurrence interval) floods to changes in 10 aquatic ecosystem services. Generally, extreme floods negatively impacted aquatic ecosystem service provisioning, while small floods contributed positively. Overall, we found that flood impacts vary depending on ecosystem properties (organic matter content) and flood characteristics (magnitude).

Reconciliation as Relationship: Reframing Settler Understanding of Reconciliation in Canada
In 2015, Canada's Truth and Reconciliation Commission called upon Canadians to reconcile relationships between Settlers and Indigenous peoples in Canada. Education for reconciliation is one important element of this process. However, critical questions arise when education is undertaken by and for Settlers such as myself: Are our undertakings actually fostering reconciliation? According to whom? Drawing from reconciliation theory and decolonizing Indigenous methodologies, a reconciliation methodology is created to consider this question in the context of three reconciliation workshops for Settlers. Indigenous perspectives and pedagogies are prioritized. The emerging understandings of reconciliation as relationship and relationship as pedagogy reframe some prevailing Settler thinking about reconciliation, unmask latent assumptions linked to the colonial habits of mind and affirm the need for personal responsibility in the reconciliation relationship. The Indigenous norm of learning in-relation is found to be a powerful experience for Settlers participants offering valuable insights for reconciliation education in Canada.

From Foraging to Farming: Changing Diet-Breadth and the Middle to Late Woodland Transition in Southcentral Ontario (ca. 1450–650 B.P.)
This study examines foraging strategies during the Middle Woodland Period's Sandbanks Phase (A.D. 700–1000) on Boyd Island, Pigeon Lake, Ontario. The faunal remains analyzed in this study were recovered from a site associated with the procurement of aquatic and terrestrial taxa. Detailed taphonomic analyses have revealed that the Boyd Island faunal remains were affected by weathering and human transport decisions. White-tailed deer was the most frequently acquired prey at Boyd Island, followed by black bear. Using the central place forager prey choice model as a framework, the analysis of diet breadth and carcass transport patterns suggests that most animal resources were acquired from both aquatic and terrestrial habitats, at moderate distances from the site. Incomplete carcasses of large game appear to have been transported away from the site, where they were subsequently processed for provisioning or consumption. Comparisons with other Sandbanks faunal assemblages and those dating to later periods indicate significant differences in terms of taxonomic composition, while continuing to emphasize the use of fish. It is suggested that the Middle Woodland foragers adopted subsistence strategies focusing on the exploitation of local habitats in which productivity may have been enhanced through niche construction associated with the low-level food production activities.
